桂芍知母汤调控cGAS-STING-NF-κB对CIA大鼠骨保护机制研究

Abstract

目的:基于cGAS-STING-NF-κB通路探讨桂枝芍药知母汤在控制胶原诱导性关节炎(CIA)大鼠炎症活动及起到骨保护作用的相关分子机制。 方法 Wistar大鼠60只,随机分为正常组、模型组、甲氨蝶呤组及桂芍知母汤低、中、高剂量组,每组10只,干预4 周,qPCR 法检测滑膜组织OPG、RNAK、RANKL mRNA 表达情况,ELISA法检测大鼠血清cGAMP、IL-6水平。结果 与正常组大鼠相比,模型组大鼠RANK、RANKL mRNA 表达上调,而OPG mRNA表达下降(P<0.05), 各治疗组与模型组相比,OPG mRNA 表达升高,RANK、RANKL mRNA 表达有所下降(P<0.05);桂芍知母汤大剂量组对OPG mRNA 上调较明显(P<0.05);随着药物浓度逐渐增加,RANK、RANKLmRNA表达抑制进一步加强(P<0.05)。模型组血清cGAMP、IL-6水平较正常组升高(P<0.05);各药物治疗组血清cGAMP、IL-6水平低于模型组,其中桂芍知母汤高剂量组改善尤为突出(P<0.05)。结论 桂枝芍药知母汤能调控cGAS-STING-NFκB轴的相关细胞因子表达,从而缓解CIA大鼠关节炎症反应,并抑制RANK/RANKL 等骨破坏相关因子的表达,起到一定的骨保护作用。
